漫长地从各种奇奇怪怪的虫终于升级到木虫了…… 纪念一下，分享点资料 preview： Cell Line: 1G1 Cell Type: mouse hybridoma (anti-chicken dorsal retina cytoplasmic membrane s, E12) DSMZ No: ACC 424 Origin: cell line was established by fusion of mouse NS-1 cells with spleen from a BALB/c mouse immunized with enriched retinal cell membranes; described as secreting a mouse monoclonal antibody (IgM) reacting with the chicken dorsal retina cytoplasmic membranes (E12) and binding outer photoreceptor elements of adult vertebrate re tina (inclusive chick, mouse, rat) References: Schlosshauer et al., Eur. Schlosshauer, Natural and Medical Science Institute (NMI ), Reutlingen, Germany DSMZ Cell Culture Data: Morphology: spheroidal cells growing singly in suspension, or occasionally loosely adherent Medium: 90% RPMI 1640 + 10% FBS + HFCS ("Hybridoma Fusion and Cloning Supplement" for the firs t 1-2 weeks post-thawing) Subculture: maintain at ca. 0.1-0.3 x 106 cells/ml; split saturated culture 1:3 to 1:10 every 2-5 days (some cells attach to the surface of the culture flask, but can be detached by tapping the flask); seed out at ca. 0.3 x 106 cells/ml; a maximal density of 1.0 x 106 cells/ml should not be exceeded Incubation: at 37 /C with 5% CO2 Doubling Time: doubling time of ca. 30 hours Harvest: cell harvest of ca. 1.0 x 106 cells/ml Storage: frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule DSMZ Scientific Data: Mycoplasma: negative in DAPI, microbiological culture, RNA hybridization, PCR assays Immunology: secretion of mouse IgM monoclonal antibody Species: confirmed as mouse with IEF of AST, LDH, NP Cytogenetics: murine flat-moded hyper tetraploid karyotype; 94-102<4n>; no centric fusion markers present Viruses: ELISA: reverse transcriptase negative |
