中药及复方含有多种复杂成分,确定效应物质基础是进行PK/PD研究的一大难题。 目前,相关研究的发展有两种趋势。一种趋势仍以寻找中药及复方中的活性物质为研究重点。与以往研究的不同之处在于,该类研究充分考虑了中药“整体观”的特点和中药有效成分在体内的变化过程,综合运用多学科知识和现代先进的分离分析手段,构建中药及其复方吸收、分布、与靶细胞结合的体内外技术平台,通过寻找中药发挥作用的体内效应成分(群) ,来追溯其在中药直接提取物中的物质形式。另一种趋势伴随系统生物学和代谢组学的发展而产生。相关研究人员认为,不仅中药的原型成分和代谢产物与中药的治疗效应有关,中药与机体作用形成的新成分也在治疗中发挥作用,由这三者组成的代谢物组才是中药发挥治疗作用的物质基础。 |
