以成分群或代谢物组为效应物质基础来进行PK/PD研究,有利于从整体上把握中药及复方的代谢特点和效应机制,但是,同时也带来一个问题。成分群和代谢物组的各组成成分在体内的吸收、分布、转化、排泄的过程都具有自身的特点,如何整合这些复杂数据得到药物的代谢动力学特征、并与效应指标形成关联,成为摆在研究人员面前的新难题。
近年来在PK/PD研究中得到应用的人工神经网络为解决以上问题提供了可能。这种计算体系不需要先假定一个特定的模型,而只需从提供给它们的数据中建立输入与输出的关系,极大地简化了传统药动学数据分析所需的建模工作,而且无论是在预示能力还是预示速度方面都有显著提高。Minor等的研究表明,用人工神经网络能够将给药情况与PD、给药情况与PK、PD与PK或其它与治疗相关的因素直接关联起来,获得它们之间的关系。人工神经网络因其出色的非线性拟合能力有望成为中药及复方PK/PD研究中一种有效的数据分析工具。
代谢组学研究常运用化学计量学理论和多元统计分析方法,对采集的海量多维原始信息进行压缩降维和归类分析,最终以较少的独立主成分综合体现原多维变量中蕴含的绝大部分整体信息(习惯上> 85% )。这种研究策略在最大化保留信息的前提下大大简化了数据分析的难度和工作量,值得中药PK/PD结合研究人员借鉴。
