[交流] α-葡萄苷酶抑制实验，阳性对照组的抑制率低，啥情况？

加入0.1mol/L、2mL的缓冲盐溶液，加待测提取液0.1mL与底物PNPG（2mmol/L,0.1mL）混合，充分振荡，37度水浴10min，加入α-葡萄糖苷酶（0.1U/mL，1mL），于37度反应20min，加入1mL，0.5mol/L碳酸钠终止反应，于400nm处测吸光度。
空白组（加酶、pnpg、碳酸钠、缓冲液）
空白对照组（pnpg、碳酸钠、缓冲液）
样品组（加酶、pnpg、碳酸钠、缓冲液、样品）
背景对照组（pnpg、碳酸钠、缓冲液、样品）
抑制率=1-（样品组-背景对照组）/（空白组-空白对照组）
1、按照样品同一浓度换算抑制率，发现阳性对照组抑制率最低。
2、用紫外测的，吸光度在0.2-0.8之间，部分样品吸光度在1左右，具体应该在什么范围才是有效的？
3、之前酶液用的是0.6ml，当加到1ml时，样品、阳性对照、空白吸光度均降低，这是怎么回事？

Inhibition (%)= (A1−A2)/A1×100,where A1 is the absorbance of control, and A2 is the absorbanceof samples. The extent of inhibition was expressed as mean ± SD(n = 3) of the enzymatic activity (IC50).
% inhibition = (A control − A sample/ A control ) × 100. Where Acontrol is the absorbance of the control reaction (containing all reagents except the test extract or quercetin) and A sample is the absorbance of the test material or quercetin. The α-amylase inhibitory activity was expressed as percentage inhibition.
