[求助] 求助：关于Brdu免疫荧光染色！！有效期至08年11月14日

我想通过BrdU结合FITC对细胞进行免疫荧光染色，同时用DAPI染细胞核，从而检测细胞增殖。实验前细胞先用含10ug/ml BrdU的培养基，37度培养2小时，然后进行染色处理，实验步骤如下：
1.Wash plates with PBS 3x.
2.Fix them in 4%PFA, 20 min at RT.
3.Wash them in PBS 3x.
4.0.5% Triton X-100 in PBS, 20min at 4℃
5.Wash slides in PBS 3x.
6.2N HCL( in Distilled water) 30min at 37℃( DNA denaturation).
7.Wash in PBS 5x
8.Incubate slides with 1%BSA in PBS at RT for 30min.
9.Rabbit anti-BrdU diluted to 1:250 with 1%BSA in PBS, incubate at 4℃ for 14-16hrs.
10.Wash with PBS 3x.
11.1: 100 dilution of FITC conjugated goat anti rabbit IgG 1%BSA in PBS, incubate at RT for 1 hr.
12.Wash in PBS 3x
13.Incubate with 500ng/ml DAPI for 8 min to stain nuclei.
14.Wash in PBS 3x.
15.Mount with 50% glycerol in PBS.

实验已经重复很多遍了，DAPI染细胞核染得很好，FITC确总也染不上。我以前从来没做过免疫染色，不知道问题出在哪，还请专家帮忙解答，我就快疯了……
