It is polysaccharides and other unknown sticky compounds. You can wash out with CTAB-free buffer (200mM Tris-HCL pH8.0, 50mM EDTA, 250mM NaCl). 1. Grind frozon tissue, add 1ml of CTAB-free buffer (cold) + 6 ul 2-mercaptoethanol. 2. Mix by inversion, keep 10 minutes on ice. 3. Spin at 10,000 g for 10minutes. 4. Discard the supernatant. (If you use particularity nasty tissue, you can repeat this step.) If it works, please give a upvote. |
