审稿人意见:
Reviewer 1: The authors identified differential expressed miRNAs in the control and irradiated liver of mice. They found 48 miRNAs were differentially expressed, including 27 known and 21 novel miRNAs. This manuscript is potentially interesting and there are several major concerns:
1. Why the authors choose liver? The liver is neither the most sensitive nor the most resistant organ to irradiation. This important information was not introduced.
2. Why the  samples were obtained 14 days after irradiation ? Why the irradiation dose was choosed?
3. Whether the dysregulated miRNAs remains up-/down-regulated in other IR doses or timepoints?
4. Although the authors predicted the potential targets of the dysregulated miRNAs, the function of the miRNAs and thier targets should be confirmed by experiments.

Reviewer 2: This report investigates effects of ionizing radiation (IR) on expression of microRNAs (miRNAs) in mouse liver tissues and identified some miRNAs were differentially regulated in response to IR. Some new miRNAs were identified. The main concern I have for the manuscript is it lacks key parameters for reviewers to judge the quality of the work. For instance, there is no information on how many animals were used, how many were treated with IR, and how many samples were processed for sequening in the study. It is hard to judge this body of potentially interesting data without these basics

问:因为深度测序贵,没钱做那么多不同时间点的和不同剂量的样品,这要怎么回复?要求加靶基因验证实验目前也做不到!因为我们是研究抗辐射的,所以没有把重心放在辐射方面!
