Oxaloacetate (OAA; 20 mM OAA, 50 mM Tris, pH 8.0) was added to the homogenate at a final concentration of 0.47 mM and the assay was initiated by addition of 200 uL CS batch solution (0.1 mM 5,5-dithiobis-2-nitrobenzoic acid in 95% EtOH, 0.15 mM Acetyl CoA,50 mM Tris, pH 8.0) for a final assay volume of 215 uL. CoA(SH) +DNTB complex formation detection was performed at 412 nm for 5 min. Background deacetylase activity was determined at 412 nm using a no-OAA control well and was subtracted from experimental wells. |
