The proper way to calibrate FA data is to use an external standard with all the analytes at equal or at least well known concentrations, and to develop response factors for each analyte that is to be applied to each and every chromatogram. Response factors vary among analytes because of injection (differential loading on the column) and detector response, and in MS it would be due to the specific ions chosen. Also, the natural response of a mass spectrometer is to MOLES, but the natural response of an FID is to GRAMS. Over molecular masses from 14:0 to 18:0 this will make a small but easily measured difference. A description of how this was taken into account is necessary. |
