[交流] Reject & Resubmit还有机会吗

Many reports on the pharmacokinetics of SHL have been reported. The paper focus on the change of Phenolic acids of Lonicerae Japonicae Flos and flavonoids of Scutellariae Radix between the single Lonicerae Japonicae Flos, Scutellariae Radix and the combination. However, the preparation of SHL is just the simple mix of the two drugs which is different from the real clinical application.
And the ration of Lonicerae Japonicae Flos, Scutellariae Radix and Forsythiae Fructus is 1:1:2. Forsythiae Fructus is a main ingredient for antiviral and antipyretic effect in SHL. However, the pharmacokinetics of Forsythiae Fructus isn’t investigated in the paper. It should be added in the experiment design.
Moreover, phenolic acids of Lonicerae Japonicae Flos and flavonoids of Scutellariae Radix contain much components than the paper reported. If the chosen ingredient can represent the characteristics of SHL?
Therefore, I have to reject the paper because the initial design. And the language still have to be revised for better understanding. For example, Shuang-Huang- Lian as a prescription is widely used in many dosage forms. The decoction is used in the paper, it should be revised in the title to make it clear.

Language need improvements

Material Methods
1. Why deuterated compounds was not used as an internal standard.
2. Autosampler was maintained at 10ºC, is it OK?
3. Preparation of samples. The samples were kept overnight in ethanolic solution, did stability of the constituents were studied under these conditions?
4. Page 6 tinidazole was used as an internal standard and on page 9, it is mentioned that the external standard method was used?
5. Page 10, It stated that internal standard was used in case of LLOQ determination, please add information about the IS, when it was added and under MRM which ions were selected?
6. The robustness of the methods was not determined?
7. P-14, 10 ml/Kg is equivalent to how much solid extract?
8. Rats were fasted for 36 hours?
9. Change the heading from Statistical Analysis to Pharmacokinetics data Analysis
10. In case of oroxyline A-7-o-glucuronide the LLOQ was 2.1 ng/ml and range was 1.61 ng/ml, how curve was constructed in the range below the LLOQ?
11. The percent recovery in some samples is very high above 109%, and %RSD is also very high, what could be the possible reason?
12. In Pk data please also add Vd/F
