三亲本杂交 荧光标记质粒

看到一片文献，利用三亲本杂交对质粒荧光标记，可是不理解：将质粒pZP201-gfp转移到Bacillus latersporus DSP (质粒pDOC)中,就能将gfp片段连接到质粒pDOC上，形成pDOC-gfp? 原理是什么? 这种方法（三亲）与用酶切克隆gfp标记有啥异同？怎么能保证成功？有没有相关文献可以借鉴? donor :E.Coli DH5α (质粒pZP201-gfp) help: E.Coli HB101((质粒 prk2013) recipient: Bacillus latersporus DSP (质粒pDOC) The insertion of gfp into the pDOC plasmid was performed by tri- parental mating in which the Escherichia coli HB101 helper strain was used to mobilize the gfp segment from the Escherichia coli DH5a donor strain into the DSP recipient strain. <Plasmid-mediated bioaugmentation for the degradation of chlorpyrifos in soil> |
