Upon grafting RGD peptides to the
PHBV-PEGMAL surface, the adsorbed amount of serum decreases
to 73(6ng/cm2 on the PHBV-RGDsurface becauseRGDpeptides
bind to the activemaleimide groups. The amount of adsorbed serum
decreases by 50% compared with that on the unmodified surface.
Moreover, the fact that PEG chains with a molecular weight of
2.3 kDa have a better protein resistance in our system than those with
a molecular weight of 5 kDa, likely because of the larger steric
hindrance from longer chains, suggests that the choice of bifunctional
PEG with a molecular weight of ∼2 kD should be more suitable in
our system. The above results indicate that our modification method
can effectively reduce the nonspecific protein adsorption from serum,
which is favorably applied in tissue engineering.

通过结合到活化马来酰胺基团导入RGD小肽到PHBV-PEGMAL表面之后,血清蛋白吸附量降低至73(单位, 6ng/cm2),比未修饰PHBV表面血清蛋白吸附量减少一半。再者,可能是由于高分子量PEG的空间位阻较高的缘故,在我们体系中分子量为2.3kDa的PEG比分子量为5kDa的PEG更具蛋白排斥效果,选择MW大约2KD的PEG更合适。上述结果表明我们的修饰方法可以有效地减少血清蛋白的非专一吸附,可以适用于组织工程应用中。
