When maleimideterminated
PEG is replaced by methoxy-terminated PEG, the
PHBV-PEGOCH3 surfaces are produced. The adsorbed serum
amount is even lower, with 32 ( 2 ng/cm2 on PHBV-PEG-
(2.3k)OCH3 and 83 ( 4 ng/cm2 on PHBV-PEG(5k)OCH3
surfaces, respectively, suggesting it is the terminal maleimide groups
that attract additional proteins, likely through the reaction with
sulfhydryls present in serum proteins at a neutral pH level. This clue
helps to explain the phenomenon observed in cell culture experiments,
that is, why cell growth is not suppressed on the PHBVPEGMAL
surface. It is clear that under cell culture conditions (37 C
and long contact with proteins in the culture medium),more proteins
could chemically bind to the terminal maleimide groups, which in
turn promotes cell growth.

用甲醚基-PEG代替马来酰胺端基修饰的PEG, 形成了甲醚基-PEG覆盖的表面。血清蛋白吸附量更低,  PHBV-PEG-(2.3k)OCH3 修饰后的表面吸附量分别是32 (单位,2 ng/cm2 ) 和 PHBV-PEG(5k)OCH3表面是8232 ( 4 ng/cm2),表明马来酰胺端基可以吸引血清蛋白吸附,有可能通过中性pH条件下与血清蛋白中巯基反应加以实现。 这个现象可以解释细胞培养实验中为什么用马来酰胺端基-PEG修饰的表面对细胞生长没有抑制作用。很清楚在细胞培养条件下(37 C和长时间接触培养基中的蛋白质)马来酰胺端基-PEG修饰的表面可以通过化学结合更多的蛋白质,导致促进细胞生长。
