摘要:为深入研究α干扰素在猪体内的调节和功能,本研究以猪为实验动物,通过RT-PCR扩增得到α干扰素基因全长编码区序列,提交GenBank数据库,获得登录号为“JQ839262”。生物信息学分析显示,该基因编码189个氨基酸,预测蛋白分子量为21.32 kD,等电点为6.67。蛋白质结构分析得出,IFN-α存在1个保守域,无跨膜结构,存在信号肽。疏水性分析显示其N端疏水,C端亲水。亚细胞定位显示,该蛋白分泌到细胞周质的概率为92.2%。系统进化分析表明,猪与牛、山羊的亲缘关系较近。同时通过荧光定量PCR法检测了α干扰素基因mRNA在猪13个组织中的表达量,发现在皮肤、肺、肌肉、心、脾中表达量较高。本研究为进一步研究猪α干扰素基因的表达调控奠定研究基础。 Abstract: For studying the regulation and function of interferon alpha in pigs, using the pig as experimental animal,and got the coding region sequence of IFN-α by RT-PCR, and to submit accession number is “JQ839262”. Bioinformatics analysis showd that the IFN-α gene encoded a protein of 189 amino acids with a predicted molecular weight of 21.32 kDa and the isoelectric point is 6.67. Protein structure analysis showed that IFN-α protein contained one conserved domain, no transmembrane regions and have signal peptide. Hydrophobicity analysis indicate that the N-terminal of the protein is hydrophobicity and C-terminal is hydrophilic. The probability of secreteing into periplasmic is 92.2%. Phylogenetic analysis showed that the pig has a closest relationship with cattle and goat .We tested the IFN-α mRNA expression in 13 tissue by qPCR, the result showed that the expression is high in skin, lung, muscle, heart, spleen. This study provide experimental basis for further study on the expression and regulation of alpha interferon gene. |
