We conducted expression analysis on the flavonoid synthetic pathway related structural genes and regulation factors by real-time fluorescence quantitative PCR (qPCR) assay based on various tissue organs (e.g., roots, stems, leaves, blossoms and seed coats) of Brassica rapa L. (Ya'an Yellow Rape) 10 d after blossoming, as well as seed coats 10 d, 20 d, 30 d, 40 d, 50 d, 60 d and 70 d after blossoming (10 d after being harvested). Our findings include: (1) The total expression of flavonoid pathway genes is the highest in blossoms while the lowest in roots and stems, and there is no significant expression difference between yellow and brown seeds. Besides, the total expression in seed coats reaches its maximum 10 d after blossoming, and it is significantly higher in brown seeds than in yellow seeds. (2) The expression of flavonoid synthetic pathway related genes in yellow seeds decreases during the whole developmental stages of seed coats, and the decreasing rate is fairly high during the early and middle stages while steady during the late stage. Moreover, the expression in brown seeds decreases steadily during the early and middle stages while increases rapidly from 50 d after blossoming (i.e., seed mature stage). There is a significant expression difference between yellow and brown seeds 20 d, 60 d and 70 d after blossoming, and this difference gets increasing with the increase of time. (3) The expression difference between yellow and brown seeds concentrates on BraANR (LDOX) and BraANS (BAN) genes during the whole developmental stages of seed coats. More concretely, the expression of BraANR (LDOX) gene in brown seeds is 38.14, 49.93 or 81.58 times that in yellow seeds 20 d, 60 d or 70 d after blossoming, respectively. We conclude that, with these observations, there is a synergistic effect among all flavonoid synthetic pathway related genes on the color difference of brown and yellow seeds, while BraANR (LDOX) and BraANS (BAN) genes are the most prominent in this synergistic effect. |
