The healthy SD rats were anesthetized with 10% chloral hydrate by intraperitoneal injection (0.3 ~ 0.4 mL / 100 g body weight). Rats were then fixed in the prone position on the three-dimensional positioning system. After sterilization of the skin at the top area of the head, the sagittal suture and coronal suture were exposed by scalp incision. A borehole with a diameter of about 1 mm was drilled in the skull 0.5 mm before and 3.0 mm next to anterior fontanelle. In experimental group, 5 μL cell suspension was injected slowly to one side of the cerebral cortex (2 mm from dural surface) with a microinjector through the borehole. In the control group, 5 μL PBS was injected slowly as described above. The microinjector was removed slowly and carefully after being retained for 5 min. The scalp was stitched and gentamicin (2000 U) was administered by intraperitoneal injection for prevention of infection. Rats were put back in their cages for feeding. |
