光谱法检测到的蛋白问题，有了答案，公示中。。

大家好，我分别用同步荧光法和红外检测了药物对人血清白蛋白的作用（该蛋白质有一个色氨酸残基） 结果如下：1同步荧光：检测到蛋白中的色氨酸残基微环境没有发生变化， 2.红外：检测到蛋白的各种形式的二级结构含量发生了变化 我的问题是：1.同步荧光检测到那个结果应该归属于蛋白的几级结构？ 2.同步荧光的结果和红外的结果矛盾吗或二者如何吻合在一起？ 多谢大家！ 答案：1.同步荧光检测到的是蛋白中的一个氨基酸残基（色氨酸），不能具体归属于蛋白的几级结构，只能说它能到影响到蛋白的空间结构（即构象）。所以，两种结果：a.如果色氨酸的微环境发生改变，则说明蛋白构象发生变化。b.如果色氨酸微环境没有发生变化，则蛋白构象不一定不变，这时实验结果对整个蛋白构象是否变化不能提供佐证。 2.红外的结果表明药物对蛋白的二级结构含量造成了影响，表明药物对蛋白的构象产生影响，即加入药物以后，蛋白的构象发生了变化。 以上为我咨询生化专业同学得到的答案，您觉的这个答案合理吗，欢迎大家批评指正，提出您的宝贵意见！ [search]蛋白[/search] [ Last edited by dnp on 2008-3-4 at 09:36 ] |

The tryptophan is a probe to indicate the conformation changes of proteins. For instance, the peak shift and intensity decrease of the fluorescence spectrum suggests the Tryptophan residue be more exposure to the polar environment. However, the reason causing this exposure can be the disruption of secondary structure or just local conformation changes. Through CD, it can analyze secondary structure change from observing changes in the range from 190nm to 220nm, while it can also reflect Tryptophan residue envrionment change via analyzing changes near 280 nm. If those two range change is cooperative, it is a proof that secondary structure change induced Tryptophan enviroment change, but not necessarily. For FTIR, through peak fit for the amide I or II band near 1650 cm-1 and 1550 cm-1, you can get the relatively percents of all secondary structure, quantitatively. |
