中文摘要         滥用肉骨粉等动物源性饲料是"疯牛病"传播的主要途径。为落实国家有关"疯牛病"防范的管理规定,满足饲料安全监管的迫切需要,项目采集了大量具有代表性的反刍动物精料补充料、鱼粉和猪、禽、牛、羊源性肉骨粉样品,按照随机区组设计法,通过在样品中人为掺入不同比例的肉骨粉,制成分析样本;利用光栅和傅立叶变换两类不同近红外光谱仪,对样本进行近红外光谱扫描测量。研究分析了肉骨粉、鱼粉和精料补充料的近红外光谱特征,揭示了基于近红外光谱的肉骨粉快速检测机理;在研究确定最优多元校正方法、光谱预处理方法以及肉骨粉检测敏感频带等的基础上,研究建立了基于近红外光谱的鱼粉和反刍动物精料补充料中肉骨粉、肉骨粉中牛羊源等不同种属肉骨粉的定性和定量快速检测方法和定标模型。结果显示,鱼粉和反刍动物精料补充料中肉骨粉定性判别的正确率分别>85%和>90%,定量检测相对分析误差RPD均>3;猪鸡MBM中牛羊源性成分的定性判断正确率>90%;肉骨粉中牛羊、鸡、猪、牛、羊等不同种属成分定量检测相对分析误差RPD分别为3.32、4.18、4.26、2.18和1.85。项目成果对于丰富我国肉骨粉快速检测手段、确保饲料安全具有重要意义。
英文摘要        The safe and healthy use of meat and bone meal (MBM) in animal feeds is of public concern in order to prevent the spread of bovine spongiform encephalopathy (BSE).The project aims at developing new methodologies for the quick detection and quantification of accidental or illegal addition of MBM in ruminant concentrate supplement, fishmeal, and of ruminant species in MBM. A large number of samples of ruminant concentrate supplement, fishmeal and MBM with diferent speciecs, pig, chicken, cattle, sheep/goat etc., were collected. Contaminated samples were got through artificial adulteration of different levels and /or different species of MBM in pure samples by means of random blocks experimental design. A Foss Scanning Monochromator NIR and a Fourier Transform NIR system were used for spectra collection. In the project, the typical spectra characteristics of diffrent species MBM, fishmeal and ruminant concentrat suplements were analysed and distinguished, which are very helpful for the recognization of the anaylitical mechanism of NIRS. Besed on the results of comparation and analysis of different spectra pretreatment methods,calibration methods and sensitive spectra regions selection results, the optimized NIRS calibartion models were developed for the quick detection and quantification of accidental or illegal addition of MBM in ruminant concentrate supplement, fishmeal, and of ruminant species in MBM. For detecting MBM in fishmeal and ruminant concentrate supplement, the correct rates of discriminant analysis were >85% and >90%, and the ratios of the standard deviation of the validation set to the standard error of prediction (RPD) of quantitative analysis were all over 3,respectively; For detecting the aduteration of ruminant in MBM, the correct rate of discriminant analysis was over 90%; For the different species of ruminant, chicken, pig, cattle and sheep/goat in MBM, the RPD values of quantitative analysis were 3.32, 4.18, 4.26, 2.18 and 1.85, respectively. These results confirm that NIRS could provide a rapid, non-destructive and non-invasive technique for the detection and quantification of MBM. The achievements of the project will play an important role in enriching MBM quick detecting methods, ensuring feed safety and preventing the spread of the BSE.
