We also tried using Tricine-SDS-PAGE to detect the occurrence of the proteolytic cleavage . Due to the greater MBP fusion protein and the very small target protein , we can see target protein with huge sample volume and can not determine whether the protein is completely digested . So we reduced the sample size to determine whether the proteolytic cleavage is complete.
