The authors show that the sensor is selective by challenging the sensor with other types of bacteria. However, E. coli O157:H7 is markedly different than these bacteria. What would happen if the sensor was challenged with bacteria more closely resembling O157-H7, such as other enterohemorrhagic Escherichia coli (O26, 45, 91, etc etc?) The authors could use a few controls demonstrating the sensor’s performance if certain modifications aren’t provided. For instance, non-specific binding is always an issue with these types of bead assays. Will GOx nonspontaneously adsorb onto the beads – i.e. without the avidin coupling? The authors take great pains to demonstrate that they are the first to use laccase in biosensing (p.8 line 20 – first report using laccase). Is this actually true? For instance, others have utilized these enzymes in biosensing: (Analytica Chimica Acta, 187, 39-45). This might be true in this sense, but the authors need to be careful in making these types of claims. As discussed previously, the optimization studies can be moved to supplemental information section, or the authors can provide greater analysis/discussion as to why these parameters were optimal. |
