Proline dehydrogenase gene is located at chromosome 22q11 deletion region ,and it is considered as the candidate genes of Schizophrenia. 脯氨酸脱氢酶 ( proline dehydrogenase，PRODH)基因位于染色体 22q11 缺失区，被认为是精神分裂症的位置候选基因。 Therefore, to explore the association of PRODH － 1945 ( T/C) polymorphism with schizophrenia in Zhuang and Han ethnic groups in Guangxi Zhuang Autonomous Region ( GZAR),of 282 patients with schizophrenia and 282 healthy controls were genotyped for PRODH － 1945 ( T / C) polymorphism by quantitative real － timePCR using TaqMan － MGB probe technology. 为此，本研究对 282 例壮族、汉族精神分裂症患者和 282 例壮族、汉族健康对照者，采用 TaqMan MGB 探针等位基因分型技术对 PRODH － 1945 ( T/C) 多态性 ( 即 rs372055) 进行基因分析，探讨该位点与广西壮族、汉族精神分裂症易感性的关联性。 1.1.1case group including Zhuang and Han ethnic groups , the inclusion and exclusion criteria of the identified studies as follows  1) the inclusion criteria: ①To keep the homogeneity of the cases , the diagnoses of schizophrenia according to ICD － 10 criteria were confirmed by 2 senior psychiatric specialists,who were strictly trained , using the consistency check of diagnostic test ; 1. 1. 1 病例组 包括壮族病例组和汉族病例组，纳入与排除标准如下: ( 1) 纳入标准: ①所有患者经至少两名副主任医师以上职称的精神科专科医师根据国际疾病及相关健康问题分类第 10 版 ( ICD －10) 精神与行为障碍分类临床描述与诊断要点确诊为精神分裂症患者，并对实施诊断的医师经过严格培训，进行诊断一致性检验，保证入组对象的同质性; 因严重兴奋、冲动不配合者，或者有严重自杀、自伤倾向者。（中译英） 上述 564 例受试者之间均无上代族间通婚史，也无亲缘关系。。（中译英） ②patient self (or their immediate families) reported patients in three generation tof family descended from the zhuang population.③patient self (or their immediate families) reported patients is Guangxi native ②患者自我 ( 或其直系家属) 报告患者三代内均为本民族血缘; ③患者自我 ( 或其直系家属) 报告患者籍贯为广西人。 1.2 genotype we put the participants’peripheral venous blood(5ml) samples into Vacuum Blood Collection Tube with EDTA, and mixed and Stored in 4℃,then extracted Genomic DNA (samples) within 3 days 1. 2 基因分型 采集每例受试者的外周静脉血 5 ml，置于含有 EDTA 抗凝剂的真空采血管内，混匀，4 ℃ 保存，并于 3 d内提取基因组 DNA。 According to the product directions,Genomic DNA were strictly operate to extract the blood by using blood Genomic DNA kit produced by TianGen technology(Beijing) Company Limited〔TIANamp Blood DNA Kit ,Catalog Number DP318DNA〕 采用天根科技 ( 北京) 有限公司生产的血液基因组 DNA 提取试剂盒 〔TIANamp Blood DNA Kit ( 离心柱形) ，目录号 DP318DNA〕，血液基因组 DNA 的提取严格按照产品说明书操作。 |
