最近看一篇英文文献，其中几句老是搞不懂，希望各位虫友帮忙看看（蓝色部分），谢谢！ The use of solid phase synthesis has two main advantages: (i) it opens the way to design new classes of promising platinum complexes which are not efficiently available by other synthetic techniques, and (ii) this new approach can be adopted for the (fast) preparation of libraries of platinum-based complexes. In this context, coordination of platinum(II) to single stranded oligonucleotides via solid phase synthesis was reported by van Boom, Reedijk, Lippert, and coworkers. This allowed for the first time an access to monofunctionally platinum(II) modified oligomers with high selectivity to complementary sequences within DNA and RNA (antigene-, antisense strategy). Sequence specific crosslinking with a target oligonucleotide could be demonstrated. During the optimization process of solid phase synthesis on-resin-compound analysis has been applied by gel phase 195Pt NMR spectroscopy. Due to the high tendency of the aminoalcohol platinum(II) complexes to form ring-closed species even at low pH values and with respect to regulatory requirements, the doubly ring closed aminoalcoholato species were selectively synthesized. |
