3.3 球形药内部孔隙的孔径均匀性控制方法
3.3.1 乳化发泡阶段改善孔结构的途径
通过在各个工艺环节对物料进行取样和结构分析,揭示出了影响成品颗粒中孔结构的影响因素,以及孔结构的形成和发展过程。其中,乳化发泡阶段的分散均匀性和影响最终孔结构的重要因素。由于该阶段是一个通常的乳化分散过程,分散相的分散程度和均匀性主要决定于剪切强度和表面活性剂的作用。通过在乳化发泡过程优化表面活性剂的品种和用量,得到了分散均匀性较高的高分子溶胶,经过分散成球和溶剂驱除后得到了孔隙直径分布均匀的样品。典型的孔结构分布情况如图9所示,通过控制乳化发泡阶段的分散强度和乳液的稳定性可以将内部孔结构的尺度控制到微米级水平。
3.3.2  分散成球阶段改善孔结构的途径
在分散成球过程连续取样,对其内部孔结构分析表明,高分子乳液的二次分散对其内部孔结构基本没有影响。在分散成球阶段只要保持相对稳定的剪切力,就可以保证球形颗粒内部结构不被破坏。
3.3.3  溶剂驱除阶段改善孔结构的途径
在该阶段影响孔结构均匀性的主要因素包括溶剂驱除的方式和溶剂驱除速度。当溶剂去除速度偏快或在较高的真空度条件下进行驱溶时,典型的颗粒孔结构分布情况如图10所示,造成内部孔隙不均匀或孔隙合并的原因在于颗粒内部的溶剂发生了快速气化。当溶剂驱除时间延长到3小时以上,并且控制升温速度平稳情况下,可以保证前期形成的孔隙结构完整地保留下来。
