Uncontrolled bleeding can also occur in the setting of an overdose of the anticoagulant heparin, leading to a need for compounds with anti-heparin activity. A novel approach to this problem takes advantage of engineered virus-like particles derived from bacteriophage Qβ (Udit et al. 2009). By generating point mutations to the coat protein and covalent attachment of poly-arginine peptides, the surface charge of the virus-like particles could be widely varied since the capsid consists of 180 coat proteins. Using an activated partial thrombin time clotting assay, they identifi ed modifi ed particles that inhibited heparin-mediated anticoagulation by > 95%. The inhibition is believed to be by presentation of regions with high positive charge density to bind heparin’s extensively sulfated domains |

肝素过量时也可出现无法控制的大量出血，这使得合成具有肝素活性的化合物成为临床需求之一。为此，2009年Udit等设计了一种由噬菌体Qβ分离出的病毒样颗粒，在其外壳蛋白上诱导点突变并共价连接聚精氨酸肽。该病毒样颗粒的衣壳由180种外壳蛋白组成，这使得其表面电荷的变化范围较为广泛。通过测定活化部分凝血酶凝血时间（APTT），作者发现这种经过修饰的病毒样颗粒对肝素介导的抗凝作用的抑制效果超过95%。他们认为其机制可能是该颗粒表面具有高正电荷密度的区域结合了肝素泛硫酸化域，从而导致抑制肝素抗凝血的效果。 本人专业并非材料，而是临床医学，基础医学知识现在也还得差不多了。。只是看到止血材料，就点进来了。材料相关的部分翻译如有不精确之处请LZ谅解。 |
