Tumor-bearing mice were injected i.v. with 2 mg pimonidazole (Hypoxylprobe-1) in 0.9% saline, 100 A g biotinylated tomato lectin (Vector Laboratories), or 0.6 mg Hoechst 33342 in PBS 30 min,10 min, or 1 min before sacrifice of the mouse, respectively. Paraffinembedded tissue blocks from formalin-fixed tumor samples were sectioned,dewaxed, and rehydrated using standard techniques. For immunofluores-cent staining, 7-μm cryo-sections were fixed in acetone for 10 min and dried in air for 1h. |

| 分别于处死小鼠前30、10和1分钟，将2 mg外源性生物还原低氧标记物哌莫硝唑(溶于0.9%盐水)、100 mg生物素标记的番茄凝集素(Vector Laboratories公司)和0.6 mg DNA染料Hoechst 33342(溶于PBS)经静脉注射到载瘤小鼠体内。从福尔马林固定的肿瘤标本中分离出来的组织块，经石蜡包埋后，按标准操作步骤切片、脱蜡、水化。进行免疫组化染色时，7 μm冰冻切片用丙酮固定10分钟，然后空气干燥1小时。 |
