9.7.2 Determination of sample size (第1段) Twenty-three completed patients in each treatment sequence (46 patients in total) would have 90% power to detect a difference in median total seizure frequency between treatments of 30% assuming a common standard deviation of 60% using a two sample t-test with a 5% two-sided significance level. A target sample size of sixty (30 patients in each treatment sequence) was chosen, based on the assumption that approximately 23% of the randomized patients might withdraw prior to the end of the study. Patients who withdrew from the study were not replaced. It should be noted that the sample size calculation was based on the percentage change from baseline, which was a secondary endpoint for this study, rather than the total seizure frequency which was the primary endpoint. This was due to a lack of reliable data on the primary endpoint when the sample size calculation was performed. The sample size calculation was performed using parametric assumptions. Although there would be some loss of power in performing the intended non-parametric analysis if parametric assumptions were subsequently considered reasonable, the most conservative estimate of variability available from the literature (other epilepsy sources) was used and the required sample size was, therefore, cautiously estimated9. The calculated sample size was estimated using nQuery Advisor version 4.0. |
