Yours sincerely Reviewers' comments: Reviewer #2: It is an interesting study on the antidepressant effect of XXX. The authors have provided some convincing data but the following revisions are needed. 1. More behavioral testing on the effect of XXX on depression like behavior casued by chroinic mild stress should be conducted. In addtion to sucrose intake test, forced swimming test and tail suspension test need to be performed. This will make the data much more convincing. 2. Why did the authors use two species for the study? They should provide the rationale for this. minor points: Annotation should be added in figure legends (Fig 3 & 4) to indicate meaning of asterisk etc. and the significant level of the data. Since the etiology of major depressionis is still unclear, discussion on differnt hypothesis of depression (e.g. neurotriphin hypothesis or neurogenesis hypothesis ) and the possible relationship between the hypotheses and the effect of XXX will provide insight for the potential mechanism of this Chinese medicine. Reviewer #3: The antidepressant-like effects of XXX was shown since it reduced immobility time in the forced swimming test (FST) and Tail suspension test (TST) without affecting locomotor activity in female mice and normalized chronic mild stress (CMS)-induced inhibition of sucrose intake in female rats. In addition, XXX also decreased the levels of plasma adrenocorticotrophic hormone and serum corticosterone and adrenal gland weight in CMS-treated rats. Overall the results are interesting, but several points diminish my enthusiasm for the manuscript. Major comments: How the doses were chosen? What is the ethnopharmacological implication? The impact of the manuscript would be enhanced if some the authors perform some additional experiments in order to propose some of the possible mechanisms underlying the antidepressant-like effects of XXX. There are some groups missing in the CMS protocol: XXX 20, 40 and 80 mg/kg and fluoxetine in nonstressed rats. Without these groups the conclusions are compromised. The whole statistical results should be provided in the Results section (F and p values and degress of freedom). Chemical composition of XXX should be provided. Minor comments: "antidepressant effects" should be changed to "antidepressant-like effects" throghout the manuscript, since the test used to assess the antidepressant activity is a predictive test of antidepressant activity. Page 10, first line: "improve the immobility time in female mice in FST and TST" - this sentence should be reframed. Page 11, willner: please change to Willner. Did you do the experiments at the same phase of the estral cycle? This needs to be specified because the phase might be creating an influence (a confounding effect) on the results. The use of female mice and rats is an interesting aspect of the manuscript considering the ethnopharmacological relevance of the work. However, I would like to know if a similar result is obtained in male rodents. Usually the immobility time in the TST is registered during the whole 6 min-session. Fluoxetine should be administered by the oral route as XXX. In the last bar of Fig 5A, 5B and 6 the correct is CMS + FLU. The references are not in the Journal Standards. |
