我查的维普的资料 干血斑样本用于HIV-1基因型耐药性检测的研究 摘要 目的研究干血斑（Dried blood spots，DBS）样本用于人类免疫缺陷病毒Ⅰ型（HIV-1）耐药性检测的可行性。方法采集受检者外周静脉抗凝血，制备干血斑样本，提取病毒RNA，逆转录套式聚合酶链反应（RT-nestedPCR）扩增HIV pol区基因序列，应用Bioedit软件分析整理后，提交theStanfordsoftware进行耐药序列分析及亚型分析。用Bayer 340bDNA核酸定量分析仪测定血浆病毒载量（VL）。结果血浆VL〉1000拷贝／ml的16例DBS和血浆（PLA）样本，扩增阳性率均为100％；血浆VL〈1000拷贝／ml的8例治疗病人的PLA样本，扩增阳性率为25％（2／8）；而DBS样本扩增阳性率为0（0／8）。DBS和PLA样本用于亚型分析结果-致，11例样本均为AE亚型。耐药性分析结果，除了极个别其它突变位点有差异外，所有的蛋白酶抑制剂主要及次要耐药相关突变，以及核苷类逆转录酶抑制剂（NRTI）耐药突变和非核苷类逆转录酶抑制剂（NNRTI）耐药突变完全相同。7例治疗病人中，对NRTI（6／7）和NNRTI（5／7）耐药率高，而且为多重耐药。结论干血斑作为-种易采集、保存条件低、便于运输的血样本，可以替代静脉采血，用于在-些实验设备条件略差的偏远地区，以及难采血病人的耐药性监测和亚型分析 |

如下是我看的文献的原文： Dried blood spots Dried blood spots as a sample collection technique for the deter- mination of pharmacokinetics in clinical studies can be used for the validation of a quantitative bioanalytical method. This tech- nique has been around for >40 years and is an easy way of col- lecting, shipping and storing blood samples. Widely used in new-born screening and therapeutic drug monitoring for trials in remote areas, it uses blood from animals/humans spotted onto collection cards, air dried and stored/shipped desiccated at room temperature. Disks are punched out of the DBS for quantitative analysis. The DBS were made by spotting 30 mL of the respective whole blood standard onto FTA cards (Whatmann, Sanford, USA) using a calibrated pipette. The samples were dried at room tempera- ture for 3 h. The samples were stored in a sealed plastic bag containing desiccant until analysis. The systematic extraction procedure was represented in Fig. 2. |
