PhD thesis at the University of Pau, France Abstract : Some fungal and algal species can be regarded as bioindicators in metal polluted environment, especially in the aquatic environment. The objective of this doctoral thesis is the imaging of metal stress at cellular level in fungi and algae. Techniques using synchrotron radiation will be employed for the localization of metals (μSXRF) and the identification of the main ligands (XAS). As complementary technique NanoSIMS will be used. This thesis will allow the first experiment with NanoSIMS in our laboratory. The thesis work requires high level methodological developments for metal imaging at sub-cellular level. The candidate will develop strategies for miniaturized sample preparation. Key words: Metals, Bioindicators, Imaging, Speciation, Synchrotron techniques, NanoSIMS. This doctoral thesis will be performed in the context of a recently obtained excellence project (EQUIPEX MARSS) at the University of Pau where one axis of the project is the acquisition of a NanoSIMS. The thesis work has the objective to start NanoSIMS imaging at Pau University. Imaging by synchrotron based X-ray fluorescence (μXRF) and X-ray absorption (XAS, μXAS) will complete the analysis of metal localization and their chemical forms (speciation) in biological organisms (aquatic fungi and algae) which can be regarded as bioindicators for metal pollution in the environment. Preliminary results for cadmium localization in an aquatic fungus already obtained by synchrotron μXRF demonstrated the feasibility of the approach.
