Data on endogenous vitamin C synthesis obtained by the essentially more reliable isotope technique involving dilution of C-ascorbic acid in intact animals yield far lower values of endogenous ascorbate synthesis than those obtained in vitro. One of the reasons is that non-physiologically high concentrations of these data ascorbate precursor were used in the in vitro experiments. Recently, have been corrected in view of the true gulonolactone concentration in the liver and the resultin~ data showed a surprisingly good agreement with those obtained in vivo. By means of the isotope technique we have further found that in expressing the endogenous ascorbate synthesis in terms of body weight the quantity of synthesized ascorbate declines with a growing body weight of the experimental animal. For instance, if the ascorbate requirement of a human body of 70 kg were to be calculated on the basis of data about requirements of mice, the human requirement would be of the order of 9,000 mg per day. Even on the basis of the rabbit's requirements the figure would be 350 mg. It is thus ,evident that a linear extrapolation to man of data obtained on small laboratory animals leads to unduly high values. |
