牛小肠源碱性磷酸酶配制与使用的问题

我买了一瓶Sigma Prod. No. P5521的碱性磷酸酶，该产品只有0.02ml的酶。产品说明书中对溶解度和溶液稳定性有以下的描述： A clear and colorless solution is observed when this product is solubilized at a concentration of 1 mg protein/ml in deionized water. Dilute solutions of alkaline phosphatase should be made in 10 mM Tris-HCl, pH 8.0, 1-5 mM magnesium chloride, 0.1-0.2 mM zinc chloride, and 50% glycerol and stored at 2-8°C. 由于本实验室没有氯化锌，我打算直接用去离子水溶解后，分装保存在-20℃下，短期内用完。不知这样的保存方法是否可行。另外，我想问问大家看有如果水解100ug的DNA大概需要多少单位碱性磷酸酶比较合适。 |
