| 碳源是微生物培养基的重要成分之一,是维持细胞正常生命活动和新陈代谢所需的碳源和能源的重要来源。分别以葡萄糖、蔗糖、橄榄油、甲醇、棕榈酸、淀粉、甘油、乳糖和麦芽糖9种常见碳源,分别以5g/L,10g/L,20g/L的浓度下,考察对菌株生长和产酶的影响。实验表明以浓度为10g/L橄榄油为碳源时,菌株生物量和酶活较高。氮是构成微生物细胞原生质和代谢中氮素来源的营养物质,因而也是微生物生长不可缺少的原料。常见的氮源有两大类,即无机氮源和有机氮源。实验结果可以看出,当培养基中添加(NH4)2SO4和NH4Cl等无机氮源时,菌株几乎不生长和产酶,而添加40g/L的豆粉有机氮源时,酶活可达到最高。 |

Being an essential part of the microbiological culture media, carbon source is also important in maintaining the energy and that of cells which are necessary for their normal living activities and metabolism. Thereby motivated, nine common carbon sources such as glucose, sucrose, olive oil, methanol, palmitic acid, starch, glycerol, lactose and maltose were utilized at the concentrations of 5 g/L, 10 g/L and 20 g/L to investigate their influence on the mycelium growth and the enzyme generation. The results exhibit that the mycelium biomass content and the enzyme activity was relatively high when 10 g/L of olive oil was taken as the carbon source. Besides, nitrogen, which performs as the nitrogen source nutrition for the composition of the protoplasm and metabolism of microbial cells, is an indispensable raw material for the essential raw materials for the microbial growth. The nitrogen sources can be divided into two main types including inorganic and organic ones. The results show that the mycelium did not grow at all when inorganic nitrogen sources such as (NH4)2SO4 and NH4Cl have been added. However, the enzyme activity would reach maximum when 40 g/L of soybean flour as the organic nitrogen source has been added. |
