红外图谱如图2所示:(a)脂肪酶、(b)纳米银介孔碳载体、(c)固定化脂肪酶。游离脂肪酶的红外图谱中3411cm-1处的吸收峰为–NH的伸缩振动,2917 cm-1和2850 cm-1处的吸收峰为甲基或者亚甲基的伸缩振动峰,2345 cm-1处得吸收峰为C-H的伸缩振动峰,1645 cm-1处的吸收峰是C-O cm-1的伸缩振动峰,1050cm-1处得吸收峰是-O-的伸缩振动峰。对比b、c的图谱可见,在固定化后的载体上出现了脂肪酶在2917 cm-1、 2850 cm-1和2345 cm-1的特征吸收峰,表明脂肪酶已经固载在载体上;同时,脂肪酶在特征吸收峰没有改变,所以酶的二级结构没有发生改变。1050cm-1的吸收峰没有在固定化酶上体现出来,可能是由于该峰是粗酶杂质的吸收峰,未被载体吸附,所以固定化酶上没有出现相应的吸收峰。
