Our strategy for the synthesis was based around the elegant build/couple/pair (B/C/P) approach described by Nielsen and Schreiber (55). The outline of our B/C/P pathway is shown in Scheme 1. The build step involves the preparation of two types of chiral building blocks: The first contains a free amine and an
azide (“azido-amine” building blocks), whereas the second contains a carboxylic acid and an alkyne (“alkyne-acid” building blocks). Coupling of three of these building blocks via amide bond formation would furnish a range of tripeptide derivatives;this process provides the basis for stereochemical diversity. The
subsequent pair phase provides the basis for scaffold diversity and will comprise of two cyclization steps.
