Morphological features and isoenzyme characterization of permanently isolated endosymbionts from green hydra have been undescribed until now. Morphological characteristics (cell area, cell radius, chloroplast area) and isoenzyme patterns of esterase (EST), peroxidase (POX), and catalase (CAT) of aposymbiotic algae from green hydra (Cx) and from reference species Chlorella kessleri (Ck) and Chlorella vulgaris (Cv) were compared. Changeable morphology of isolated algae determined by light and electron microscopy was noticed. Results obtained by the native anodic electrophoresis suggest complex relations within the Chlorella genus. The greatest isoenzyme diversity was noted in EST patterns of isolated endosymbionts. Due to their much higher heterogeneity, EST isoenzymes seemed to be more suitable genetic markers for identification of different Chlorella species than CAT and POX isoenzymes. | 到现在为止,对从绿色水螅中分离出来的endosymbionts已经进行了形态特征研究及其同工酶的表征。对从绿色水螅中分离出来的aposymbiotic的 同工酶(酯酶,过氧化酶和过氧化氢酶)的表征及其形态特征(细胞面积、半径、叶绿素面积)与普通小球藻、特异相关性的小球藻的比较发现:分离出来的藻类的形貌发生了改变(此形貌通过光学、电子显微镜进行表征)。通过原位的阳极电泳结果显示其属于小球藻的种属一员。其中endosymbiont的EST类型的同工酶种类多样性最多。由于EST类型的同工酶具有更大的异质性特征,它比CAT\POX同工酶更能作为种属区分的遗传性特征。对所研究的酶的形态研究更加证实了同工酶分析中的种间差异。 |
