[交流] 求达人翻译两段话~~关于质谱定量蛋白的文献~~

第一段:
此方法的基本策略是:在被定量蛋白质的氨基酸序列中选择一段相对分子质量大小适中(一般为0.9~2 kd)、质谱信号响应较强的肽段作为模板,用化学合成或细胞培养的方法合成带有一定同位素(如13C、15N、2H) 标记的相同肽段(仅相对分子分量有差异) 作为内标,通过检测肽段的质谱信号强度之比(峰高或峰面积比) 来对相应蛋白质进行绝对定量。应用内标法的前提条件是:1) 被绝对定量的蛋白质酶切要充分;2) 被选择作为内标肽段模板的肽段和蛋白质间存在一定的定量关系。
第二段:
此方法的策略是:将待测定的蛋白质在强酸条件下,进行完全的水解,全部形成氨基酸混合物,然后加入已知量的同位素标记的氨基酸作为内标,经LC-MS/MS质谱检测一对氨基酸的质谱峰强度以实现对氨基酸的浓度测定,再根据此氨基酸在蛋白中的含量,计算出蛋白的量。

The basic strategy is as following: in the quantitative amino acid sequences by selecting the size of a medium molecular weight (typically 0.9 ~ 2 kd), mass spectrometry peptide strong signal response as a template, using chemical synthesis or cell culture methods synthesized with a certain isotopes (eg 13C, 15N, 2H) peptide labeled the same (only with different molecular weight) as the internal standard, detection of peptides by mass spectrometry signal intensity ratio (peak height or peak area ratio) of absolute quantification of the corresponding proteins. Application of the standard method provided that: 1) The absolute quantification of protein digestion to fully; 2)  It was chosen as the internal standard template peptide between peptides and proteins there is a certain quantitative relationship.

This method's strategy is: to be the determination of protein in acid conditions, full hydrolysis, all the formation of amino acid mixture, then add a known amount of isotope labeled amino acid as the internal standard by LC-MS/MS mass spectrometry one pair amino acids in order to achieve mass peak intensity of amino acid concentrations were measured, and then based on this amino acid in the protein content, calculate the amount of protein.
