| 核苷类药物作为目前临床上治疗艾滋病、肝炎、疱疹等病毒性疾病的主要治疗药，不仅抑制病毒复制的酶，而且能作为底物类似物参与竞争性渗合入病毒复制的DNA中，阻断DNA链的延长，从而抑制病毒复制。但存在毒副性大，需长期用药，易使患者产生耐药性等缺陷。开环核苷类化合物作为核苷类抗病毒药物的重要一族，具有低毒、低耐受性和广谱抗DNA病毒活性，对耐药菌株有强烈的杀伤作用，在抗病毒治疗领域占有重要地位，是近年研究的热点。 |

As the major therapeutic drugs for clinical treatment of AIDS, hepatitis, herpes and other viral diseases,nucleoside drugs can not only inhibit the enzyme which the viral replicate, but also penetration into the DNA which the competitive virus replicate as a substrate analogues. It interrupts the extension of DNA chain,and then restrains virus to duplicate. However, the drugs are with such defects as bad side effect ,long-term medication needs and easy to get drug resistance in patients. As an important antiviral family of nucleoside drugs,Open-loop nucleoside compounds get advantages as low toxicity, low tolerability and broad-spectrum anti-DNA virus activity.it is efficent in killing resistant strains. It plays an important role in the field of antiviral therapy and it had become the research hotspot in recent years. |
