Genetic adjuvants for DNA vaccines


Abstract
The relatively low efficacy of DNA vaccines in inducing immune responses,especially in large animal species and humans,has
impaired their practical use.Despite considerable effort expended on improving DNA vaccine delivery,only minute amounts of
Ag are available for immune induction following DNA vaccination.Two complementary strategies have been used to improve
and modulate the immune response induced by DNA vaccinesi)supplementing DNA vaccines with plasmids encoding cytokines
and(ii)targeting the Ag encoded by DNA vaccine through genetically fusing the Ag to molecules binding cell surface receptors.
This paper reviews recent progress in these two areas and possible mechanisms responsible for the observed effects.?2001

摘要
DNA疫苗表现在多数动物物种和人类上低效能的诱导免疫应答限制了它们的实际应用。尽管在提高DNA疫苗“诱导”上花费了相当努力,只有极少量的Ag在DNA疫苗的作用下发生免疫应答。过去有两种方法来提高和调节DNA疫苗诱导的免疫应答:1)补充带有细胞因子编码质粒的DNA疫苗2)通过DNA疫苗融合Ag到和细胞表面受体结合的分子来靶向编码Ag。本文综述了近年来在这两个领域的进展和可能的机制,以及研究其效果。
