如题所示,关于棉花转基因方面的!只要意思大概对即可! 由图4可知,经过卡那霉素筛选后的T0代棉花植株进行两代自交筛选后得到5个纤维长度明显变短的纯系,其中3个为E6启动子,2个为35S启动子。五个纯系纤维长度均短于对照,其中E6AS-2、35AS-1显著变短。Southern检测结果表明这5个纯系为1-4个拷贝不等,其中E6AS-1、E6AS-2为单拷贝,其余为多拷贝。定量PCR分析结果表明,反义转基因纯系中GhSUSA1基因在开花后25d被抑制最为明显,开花后10d表达量也均被显著抑制,其余时期转基因株系表达量没有对照高。其中35AS-1株系在整个时期表达量都很低。 对纤维长度、强度和酶活进行了统计分析,由表2可知这5个纯系纤维长度都不同程度的变短了,尤其是E6AS-2和35AS-1,这两个纯系在0DPA表达量就很低,在10DPA表达量也低于其他株系,因为10DPA是纤维伸长最为活跃的时期,推测纤维长度变化与10DPA表达量被显著抑制有关;同时转基因株系在10DPA的酶活也低于对照,酶活测定结果与表达量相一致,与纤维长度呈显著正相关,相关系数为r=0.853。反义转基因株系纤维强度也受到影响,5个株系强度均有所下降,尤其35AS-1下降最为明显,而35AS-1在次生壁初始形成的23DPA表达量最低。将纤维强度和23d相对表达量比较发现,二者呈较高正相关(r=0.847)。这一结果表明蔗糖合酶的抑制不仅能影响纤维伸长而且使纤维强度降低了。 35AS-1除了上述纤维品质方面的变化,成熟棉铃还较对照显著变小了。对35AS-1和35AS-2测定了铃重和百粒重,结果显示35AS-1棉铃重量显著减小,百粒重35AS-1和35AS-2两个纯系均显著减小。说明在35S启动子下,GhSUSA1基因的抑制能够影响种子质量。 正义转基因载体由于是GUS融合载体,转基因后代经GUS染色后筛选获得阳性单株。目前转基因纯系虽然没有完全获得,但就T0代纤维表型来看至少出现了3个纤维显著变长的单株,且纤维长度都在30mm以上。 |
