Two MCF-7 sub-lines MN1 and MDD2 were generously provide by Dr Galmarini (Inserm 453, Laboratoire de Cytologie Analytique, France) and cultured in DMEM media (Winsent (St Bruno, Qc)) supplemented with 10% SVF. MDD2 line derives from the transfection of MCF-7 cells with a dominant negative mutant of p53 (pCMV-DDp53; mut-p53) that codes for a non-functional p53. MN1 line is the control line transfected with the empty plasmid. Some MN1 cells were transitionally transfected with pCDNA Bcl-2 using the lipofectamin system from Invitrogen (Burlington, Ont.). The human lung carcinoma H1299 line (ATCC (Manassas, VA)) does not express p53 and was cultured in RPMI media (Winsent (St Bruno, Qc)) supplemented with 10% SVF. Human colonic carcinoma cells HCT116 and the p21 null HCT116p21K/K lines were a gift to our laboratory from Dr Vogelstein B. (John Hopkins University, Baltimore, MD), and were cultured into McCoy media (Wisent (St Bruno, Qc)), supplemented with 10% SVF. |
