Epigenetic modifications act on DNA and its packaging proteins, the histones, to regulate genome function. Manifest as the methylation of DNA and as post-translational histone modifications, these molecular flags influence the architecture, integrity and packaging of the chromosome, the accessibility of DNA to regulatory components and the ability of chromatin to interact with functional nuclear complexes. While a multicellular individual has only one genome, it has multiple epigenomes reflecting the diversity of cell types and their properties at different times of life; in healthy and in compromised situations. The relationships between the underlying DNA sequence, the dynamic changes in epigenetic states and the consequences of this for the cell or organism, are beginning to emerge as the functions of epigenetic modifications in particular model systems and model organisms are studied. These integrated approaches go hand-in-hand with genome-wide studies describing the location of epigenetic modifications in different cell types at different times that allow functional correlations to be made.

The excitement and curiosity surrounding epigenomics has resulted in a growing community of researchers in a burgeoning field and the development of new technologies built on the backbone of genome sequencing projects. This has resulted in a recognition that the adaptability and vulnerability of epigenetic states has profound effects on natural variation, the response of the genome to its environment and on health and disease.

The aim of this volume is not to describe epigenomes, but rather to explore how understanding epigenomes tells us more about how biological systems work and the challenges and approaches taken to accomplish this. These contributions have attempted to integrate epigenomics into our understanding of genomes in wider context and to communicate some of the wonders of epigenomics illustrated through examples across the biological spectrum.

在基因组的水平上研究表观遗传修饰的领域被称为"表观基因组学"。

表观基因组学使人们对基因组的认识又增加了一个新视点:对基因组而言,不仅仅是序列包含遗传信息,而且其修饰也可以记载遗传信息。   

欧洲的生物学家于1999年成立了"人类表观基因组联合研究体"。
2004年9月24日欧洲还进一步成立"表观基因组学"先进研究网络,表观基因组学研究已渐入佳境。

有关专家认为,过去教科书一直认为遗传的分子基础是核酸,生命的遗传信息储存在核酸的碱基序列上。随着"表观基因组学"的兴起和有关研究工作的全面进展,这种经典的遗传观念正面临着巨大的挑战。

不可否认,基因对生命具有非常重要的作用,基因的异常通常就会导致生命的异常。但是,作为开放的复杂系统,生命活动从来就不是完全由基因决定的。当前越来越多的证据在向"基因决定论"挑战,这其中最突出的就是表观基因组的作用,通过这一领域的最新研究,科学家正在以一种全新的视野来理解生命现象。
