An antibacterial fluoroquinolone enoxacin enhanced RNA interference (RNAi)

Although major protein components of the RNA interference (RNAi) pathway have been identified, regulatory mechanisms of gene expression for the RNAi pathway remain largely unknown.  Here, Shan et al. have reported a small molecule antibacterial fluoroquinolone enoxacin enhanced RNAi, and its basic principle.  To monitor the activity of the RNAi, they developed a stable cell line (RNAi-293-EGFP)-based green fluorescence detection system.  The cell line was prepared from human cell (293-EGFP), expressing gene encoding enhanced green fluorescent protein (EGFP), and was infected with a lentivirus expressing a short hairpin RNA (shRNA) targeting EGFP mRNA.
Using this system, they identified enoxacin increased EGFP gene knockdown mediated by siRNA against EGFP in a dose-dependent manner.  They showed that enoxacin could facilitate the interaction between trans-activation-responsive region RNA-binding protein and RNAs.

从人类细胞得到一个稳定的细胞株,该细胞株表达增强的绿色荧光蛋白(EGFP),并用一个慢性病毒感染,该病毒能表达shRNA,靶向EGFP的mRNA。这个稳定的细胞株被用来检测RNAi的活性。通过这个监测系统,证明氟啶酸能增强SiRNA介导的基因敲除,并且有剂量依赖性。氟啶酸的作用机制在于易化RNA结合蛋白与RNA的相互作用。

原意大概是这样的,就是RNAi的程度,用荧光来检测
