An antibacterial fluoroquinolone enoxacin enhanced RNA interference (RNAi) Although major protein components of the RNA interference (RNAi) pathway have been identified, regulatory mechanisms of gene expression for the RNAi pathway remain largely unknown. Here, Shan et al. have reported a small molecule antibacterial fluoroquinolone enoxacin enhanced RNAi, and its basic principle. To monitor the activity of the RNAi, they developed a stable cell line (RNAi-293-EGFP)-based green fluorescence detection system. The cell line was prepared from human cell (293-EGFP), expressing gene encoding enhanced green fluorescent protein (EGFP), and was infected with a lentivirus expressing a short hairpin RNA (shRNA) targeting EGFP mRNA. Using this system, they identified enoxacin increased EGFP gene knockdown mediated by siRNA against EGFP in a dose-dependent manner. They showed that enoxacin could facilitate the interaction between trans-activation-responsive region RNA-binding protein and RNAs. 各位学生物的,麻烦看看这段话符合反应,我学有机的,实在不能理解。太纠结了。 |
