The dried roots of S. chinensis (500 g), which were collectedon Geochang, Korea, were ground and extracted with 95% MeOH (1.5 L · 2) at room temperature for 24 h. After filtration andconcen tration, the MeOH extract (75 g) was suspended in H2O and then partitioned with the order of n-hexane, CHCl3, andEtOAc to give three fractions, n-hexane- (9.0 g), CHCl3- (17.0 g), andEtOA c-soluble residue (22.0 g), respectively. The EtOAc-soluble fraction exhibiteda ctivities against hACAT- 1 andhACAT- 2, with 81% and58% inhibition at 100 lg/mL, was subjectedto flash silica gel column chromatography with a gradient of chloroform–methanol (100:0 to 0:100, v/v) to give five fractions. By bioassay-guided fractionation, the one of active fractions was purifiedby column chromatography on silica gel with a gradient of n-hexane–ethyl acetate (3:1 to 2:1, v/v) to obtain the sesquineolignan, aucerneol B (1) (20 mg). Subsequently, the other active fraction was subjectedto column chromatography on silica gel with a gradient of n-hexane–ethyl acetate (1:1 to 1:2, v/v) to give dineolignans, manassantin A (2) (200 mg), and manassantin B (3) (1 g). |

与楼上相似，但是细节不一样 S.chinensis干燥根源于韩国Geochang,用95%的甲醇1.5升室温下各24小时提取两次。过滤后浓缩，75g甲醇提取物用水混悬后分别用正己烷，氯仿，乙酸乙酯萃取，得到提取物分别为正己烷中9g,氯仿中17g，乙酸乙酯中22g.乙酸乙酯部分表现出强 hACAT-1 和hACAT- 2抑制活性, 100lg/mL分别为81%和58%。乙酸乙酯部分采用柱层析色谱法，以氯仿-甲醇100：0-0：100梯度洗脱，得到5部分。通过生物活性追踪，有活性的一部分用硅胶柱纯化，洗脱剂为正己烷-乙酸乙酯3：1-2：1.得到 aucerneol B 20mg.随后其它活性部分采用硅胶柱层析用正己烷乙酸乙酯1：1-1：2梯度洗脱得到manassantin A 200mg 和manassantin B 1g. |
