1.Protein Nano-beads as alternative cell targeting particles for nucleic acid based vaccination and gene therapy
According to the experience of the institute with nano- to milli-meter scale protein hydrogel spheres for cell culture applications and for immobilization of pathogens for diagnostic applications, these nano-meter scale hydrogel beads are suited to function as delivery vehicles for DNA to a variety of human cell lines as measured by the uptake of Gfp-coding plasmids and subsequent transient expression and flowcytometric analyses. These particles will now be tested for the delivery of modified mRNA molecules for the expression of Gfp, protein photosensitizers and SARS-Cov 2 spike protein. Specific cell targeting will be tested via functionalization of the particles with aptamers against marker proteins in combination with click-chemistry.

2.Microbiome engineering for the Attenuation of microbial dysbiosis upon aging
The composition of the human microbiome has enormous effects on several diseases like Alzheimer, Parkinson, adipositas and on malfunctions of the intestine like Morbus crohn and Collitis ulcerosa. However, aging also is accompanied by fundamental changes of the microbial diversity in the gut and the relative composition of the microbiome. This may result in dysbiosis which in turn appear to affect the immune system. In cooperation with aging experts “lead” dysbioses will be defined which occur in aging mice and the relevant bacteria detected by next generation sequencing will be isolated and cultivated. Biotechnological processes will be developed for their reliable medium scale production to provide high quality cell material for feeding experiments. Key questions will be to address correlations of individual bacterial strains and their impact on the aging immune system, how dysbiose in the gut as a consequence of aging can be (stably) reverted and how the supplementation of the new probiotics can influence the immune system and health.

3.Azulitox-based photosensitizer/ aptamer conjugates for the development of a photodynamic therapy against prostate cancer
Azulitox is a reasonably cancer specific photosensitizer protein that can cause cell death upon illumination with blue light which has been published by our group. The directed and specific delivery to individual tumors will be optimized by equipping the protein with binding entities that bind to target specific cell surface markers. In this case these binding molecules will be alkine-modified aptamers evolved against a prostate cancer cell marker, which will be used to be conjugated to the NHS-azide  functionalized azulitox protein. In cooperation with the Institute for Laser Technologies in Medicine & Metrology (ILM) the concept will be evaluated in different tumor models and the illumination regime for close to real photodynamic therapy approaches will be optimized.
