Laboratory of RNA Biology headed by Prof. Joanna Kufel (https://kufel.igib.uw.edu.pl/) is looking for PhD student at the Institute of Genetics and Biotechnology, Faculty of Biology, University of Warsaw for the project “Mechanistic study of transcription regulation in Arabidopsis” Starting date: 01.06.2022, duration 3 years. Project description: This project involves comprehensive studies of transcription regulation in Arabidopsis thaliana by three proteins, XRN3, DXO1 and RSA1. XRN3, a nuclear 5'-3' exoribonuclease, is involved in the torpedo transcription termination mechanism of RNA polymerase II. DXO1 is an enzyme, present in the nucleus and the cytoplasm, capable of removing the non-canonical NAD+ cap on RNAs, but its major functions are unrelated to catalytic activity. In turn, RSA1 is a nuclear protein that forms liquid-like condensates that contain components of the transcription machinery and RNA-related proteins. The main objective is to dissect the exact mechanism of their joint contribution to transcription initiation, elongation and termination, possibly through condensate formation (liquid-liquid phase separation, LLPS) with the transcriptional machinery. We also want to assess the impact of transcriptional activity of these factors on cellular processes and plant physiology, including response to environmental stresses.
