一种酶是否需要多种辅酶才能发挥酶活呢？

1.各位虫友好，最近在做的酶的催化时，发现该酶不仅需要NADH，也需要FMN，请问这种酶需要这两种辅酶才能发挥酶活吗？但是从酶活测定上看，只有NADH被消耗了，那这FMN的作用是什么呢？ 2.另外为什么要选择磷酸钾缓冲液呢？磷酸钠是否也可以呢，钾钠在酶催化上游什么区别吗？ 3.以下是酶活验证步骤，请各位虫友指教了：The enzyme assay of ca2ENR was performed by monitoring the consumption of NADH using a DU® 800 UV/Visible Spectrophotometer . A reaction mixture containing 100 mM potassium phosphate (pH 7.0), 0.2 mM NADH, 5 μM FMN and 5 μM purified ca2ENR protein was prepared in a quartz cuvette with a butyl rubber stopper. After replacing the head space with nitrogen gas, a reaction was initiated by addition of 10 mM 4ACA at 30 °C. The reduction of 4ACA into 4AHCAwas measured as the decrease in absorbance at 370 nm due to oxidation of NADH. |
