作为通讯作者,第一次投稿,收到审稿意见,有些拿不准怎么回复,不知道小木虫中是否有有经验的老师帮我分析下,不胜感激 we have received the reports from our advisors on your manuscript, "***", which you submitted to ***. based on the advice received, i have decided that your manuscript requires some important revisions. when preparing your revised manuscript, you are asked to carefully consider the reviewer comments which can be found below, and submit a list of responses to the comments .please note that the figure in the supplementary material must be redrawn to a publishable standard. you are kindly requested to also check the website for possible reviewer attachment(s). while submitting, please check the filled in author data carefully and update them if applicable - they need to be complete and correct in order for the revision to be processed further. in order to submit your revised manuscript, please access the following web site: comments to the author (if any): reviewer #1: title: *** the authors have successfully examined the effects of glycerol and sorbitol on the hydration status of ***. they have also adjusted the volume fractions of glycerol or sorbitol (0-80%) to tune the water activity around the ***. preparation processes are described in detail and corresponding characterisations are carried out. however, i have some comments and suggestions to improve the overall quality of this work. 1. the authors could have studied the effect of the polyols on *** for longer time span to demonstrate the stability aspect of the protein in a foreign environment. 2. use of glycerol and sorbitol might generate in vitro o2 due to hydration. if this is true then the authors could have also looked for the effect of glycols addition on superoxide dismutase (sod) activity. 3. the charge transfer mechanism of the *** in the presence of these glycols is unclear and the authors should schematically explain the origin of charges. you can refer to the following link for better understanding. 4. there are some inconsistencies in the supplementary information. the fonts used is very blurry which makes it difficult to read and understand. reviewer #2: the authors in the current work investigated the effect of glycerol and sorbitol on the excitation dynamics of the ***. the subject is novel and interesting and it is part of the frontiers of bioenergetic investigations. the biochemical and biophysical tools used here belong to up-to-date research methods. the conclusions are mostly sound, however, as can be seen from my comments below, additional clarifications are needed before publication: 1. my major concern is the assessment (or the lack thereof) of the stability of the complex upon exposure to glycerol and sorbitol for extended period of time. fig. 2 displays spectra that were produced as the averages of the actual spectra recorded at 2 and 6 hours after solubilization. i will address later the term "solubilization" but i assume that 2 and 6 hours after addition of the polyols. glycerol, while being a great cryoprotectant, is known to greatly decrease the stability of photosynthetic complexes at room temperature. how much of the observed spectral changes on fig. 2 (decreased intensity and shifts) are associated with protein degradation due to the presence of the polyols? neither under 3.1 nor in the caption of fig.1 was mentioned whether the *** were also taken at the same time after the addition of the polyols as the optical spectra. it should be clarified if any spectral change is due to degradation and also be made clear that the spectra shown in figs. 1 and 2 were taken under the same conditions. 2. throughout the text and in figure captions the authors use the term of "solubilization" of *** either in detergent solution or in buffer solution. the *** is dispersed in detergent micelles. therefore, the terms *** in buffer solution (e.g. caption of all figs), aqueous solution (e.g. page 3, line 41), and detergent solution (e.g. abstract; page 2, line 17; page 4, line 47) are also incorrect. the samples are in dispersed, micellar system that contains multiple phases. the distribution (partition coefficients) of the polyols is not uniform in these phases. do the authors have data to address the partition coefficients of the polyols in the aqueous phase, the micellar phase, and in the protein phase. some sort of a distribution is expected as glycerol often show up even in crystal structures of photosynthetic complexes. 3. what do the solid lines represent in fig. 3? if the authors used a model and did fit the experimental data with that model, a smooth, fitted curves should be shown and identified in the figure caption. if no model was used the lines must be removed. it is inappropriate to connect the data points with straight lines. 4. figure captions are generally very frugal and as per (2) are often incorrectly describe the system used. |
