在中国南极科学考察期间,从南极海域采集了水体,沉积物等样品,从中分离得到一批嗜冷细菌,采用MTT法对155株南极海洋细菌进行了细胞毒活性筛选,其中176号菌株的抗肿瘤活性最高。通过用薄层色谱分离，确定最佳分离条件为石油醚：丙酮=8:2，并做了粗提取，发现组分5由最高的抗肿瘤活性。通过对组分5理化性质的研究发现：①紫外稳定性，活性物质的抑瘤活性随照射时间的加长而降低；②温度稳定性，活性物质的抑瘤活性随处理温度的升高而迅速下降；③pH稳定性，活性物质的抑瘤活性在pH 7-11范围内活性与对照无差异；在pH值3-7之间，活性明显下降，pH值为3时，其活性则完全丧失。 [ Last edited by whlk2000 on 2009-6-7 at 22:25 ] |

During the Chinese scientific investigation of South Pole, samples such as water and settlings from territorial waters of South Pole have been collected, from which a quantity of psychrophilebacteria were separated.The active of No.176 bacterial strain is the highest after the cell cytotoxin active substances selection of 155 isolates marine bacterium by MTT. After the separation by thin layer chromatography, the optimal conditions for separation is that the ratio of petroleum ether to acetone is 8 to 2.And Component 5 is found to have the highest anti-tumor activity after the crude extraction.The study for physicochemical property of Component 5 showed ①ultraviolet stability that anti-tumor activity of active substances is lower with longer irradiation time;②temperature stability that anti-tumor activity of active substances rapidly declines with processing temperature going up;③pH stability that that anti-tumor activity of active substances has no differences with comparation from pH7 to pH11.from pH7 to pH11, the anti-tumor activity is obviously decreasing.The activity is totally lost at pH3 |
