长期以来,恶性肿瘤对人类生命健康构成了严重威胁。药物治疗作为肿瘤治疗的主要手段,一直受到人们的重视。临床上常用的抗肿瘤药物有50多种,但大多数药物只能使病情缓解,无法达到完全治愈的目的,所以抗肿瘤新药的开发一直是药物研究领域的主要方面。
由于海洋环境十分独特,生存在这种环境下的微生物与陆地微生物相比较,它们具有一些独特的代谢途径和遗传背景,可能产生特殊代谢产物,其化学结构类型多、活性显著,是发现新活性先导化合物的重要源泉、近年从海洋来源的微生物代谢产物中发现许多结构新颖的活性物质,引起广泛的关注。
在中国南极科学考察期间,从南极海域采集了水体,沉积物等样品,从中分离得到一批嗜冷细菌,采用MTT法对150余株南极海洋细菌进行了细胞毒活性物质的筛选,其中176号菌株的抗肿瘤活性最高,本研究对抗肿瘤化合物进行了初步的分离纯化,掌握了该化合物的基本理化性质,为进一步分离纯化奠定了基础。

The malignancy is seriously threatening the human life and health for a long time. Medication as  primary tumor therapy has been rewarded much attention. There are more than 50 kinds of antitumor drug in routine clinical practice, but most of them can only ease the patients’ condition, not completely cured. Therefore, the development of new antitumor drugs has played a main role in the area of drug research.
    Due to the very unique marine environment, microorganisms surviving in the marine environment, compared to ones on land, have some unique metabolic pathways and genetic backgrounds and maybe create special metabolites. They have many types of chemical structures and obvious activity, which is the important source of the new active lead compounds. In recent years, a lot of novel active substances have been discovered in the microorganism metabolites from the ocean, which is attracting widespread attentions.
    During the Chinese scientific investigation of South Pole, samples such as water and settlings from territorial waters of South Pole have been collected, from which a quantity of psychrophilebacteria were separated. The active of No.176 bacterial strain is the highest after the cell cytotoxin active substances selection of more than 150 isolates marine bacterium by MTT. This study had preliminary separation and purification for antitumor compound and mastered basic physicochemical properties of the compound, which laid the foundation of further purification.
