| PK/PD结合模型由Sheiner等于1979年建立。自建立以来,它在化学药物理论研究及临床应用领域得到了广泛的应用,其研究内容涵盖了药物的体内代谢过程、药物与酶的对应关系和相互作用、药物毒副作用、耐受性与致敏性研究、立体异构体药物比较等众多方面。该模型在化学药物研究中的应用不仅为解释药物的代谢机制、作用机制或耐受机制等提供了信息,也为临床剂量调整、优化治疗方案、防治剂量相关性不良反应提供了依据。将PK/PD模型应用于中药研究,一方面可以加深研究人员对中药作用机制的认识,有利于以现代科学阐述中药组方原理,为研究古方、筛选新方提供科学依据和方法, 另一方面,它还可以促使中药制剂的“给药精密化”,为拟定给药方式、剂量及间隔时间提供依据,从而提高临床治疗水平、减少不良反应,对于实现中药治疗学的现代化和科学化有重要意义。PK/PD模型在化学药物研究中取得的成绩,为中药PK/PD研究提供了思路和方法学参考。已有研究者就该模型在中药领域的应用进行了探讨。 |

| PK/PD combination model was built by Sheiner et al in 1979. It has been broadly applied in reseaches of the Chemical medicine theory and clinical practice since it's building. The researches on PK/PD combination model cover the metabolic process of medicine in vivo, the correspondance and interaction between medicine and enzymes, the side-effects of medicine, tolerence and sensitivity of medicine and the comparison of stereoisomers etc。This model provide not only information for the explaning drug metabolism mechanism, therapeutic mechanism and tolerence mechanism, but also the basis for clinical dose adjustment, therapeutic scheme optimization and the prevention of to dose-related side-effects. The application of PK/PD model in Chinese medicine researches will help the researchers in understanding better the therapeutic mechanism of Chinese medicine, and exploring the prescription principle of Chinese medicine to build the scientific basis and method for studying ancient prescriptions and selecting new prescription. On the other hand, it will promot the dose accuracy of Chinese medicine and provide the basis for making protocals of administration ways, dose and intervals, and thereby improve the theapeutic effect and induce side-effect. So this model will be very meaningful to the modernization and scientification of the Chinese medicine therapeutics. The successful application of PK/PD model on researches of chemical medicine provide good references in thoughts and methodist for the PK/PD model study of Chinese medine. Some researchers have explored the application of this model in Chinese medicine field. |
